Developmental pathways associated with cancer metastasis:Notch,Wnt,and Hedgehog

摘要 Masterdevelopmentalpathways,suchasNotch,Wnt,andHedgehog,aresignalingsystemsthatcontrolproliferation,celldeath,motility,migration,andstemness.Thesesystemsarenotonlycommonlyactivatedinmanysolidtumors,wheretheydriveorcontributetocancerinitiation,butalsoinprimaryandmetastatictumordevelopment.Thereactivationofdevelopmentalpathwaysincancerstromafavorsthedevelopmentofcancerstemcellsandallowstheirmaintenance,indicatingthesesignalingpathwaysasparticularlyattractivetargetsforefficientanticancertherapies,especiallyinadvancedprimarytumorsandmetastaticcancers.Metastasisistheworstfeatureofcancerdevelopment.Thisfeatureresultsfromacascadeofeventsemergingfromthehijackingofepithelial-mesenchymaltransition,angiogenesis,migration,andinvasionbytransformingcellsandisassociatedwithpoorsurvival,drugresistance,andtumorrelapse.Inthepresentreview,wesummarizeanddiscussexperimentaldatasuggestingpivotalrolesfordevelopmentalpathwaysincancerdevelopmentandmetastasis,consideringthetherapeuticpotential.EmergingtargetedantimetastatictherapiesbasedonNotch,Wnt,andHedgehogpathwaysarealsodiscussed.
