简介:稻米蛋白质含量是水稻(OryzasativaL.)营养品质育种的重要内容之一。本研究以珍汕97与南洋占杂交构建的重组自交系(RIL)群体为材料,结合其建立了由190个SSR标记组成的遗传图谱,利用QTLmap-per1.6对糙米和精米中的蛋白质含量的遗传基础进行了分析,共定位到4个控制糙米含量的QTLs和2个控制精米含量的QTLs。其中,控制精米蛋白质含量的2个QTLs与控制糙米蛋白质含量的2个QTLs位置一致,这2个QTL(qpc1和qpc2)在糙米和精米的蛋白质含量中均解释了较大的表型变异,而且糙米和精米蛋白质含量的相关系数为0.814,这表明糙米和精米的蛋白质含量具有相似的遗传基础。研究还发现上位性在糙米和精米蛋白质含量的遗传中也起很重要的作用。其中,在糙米蛋白质含量中,上位性QTLs可以解释42.2%的表型变异;在精米蛋白质含量中,上位性QTLs共解释了27.8%的表型变异。本研究初步揭示了稻米蛋白质含量的遗传基础,为分子标记辅助选择改良稻米品质及蛋白质含量基因的克隆提供了有益的信息。
简介:QTLmappingforsevenqualitytraitswasconductedbyusing254recombinantinbredlines(RIL)derivedfromajaponica-japonicaricecrossofXiushui79/CBao.Theseventraitsinvestigatedweregrainlength(GL),grainlengthtowidthratio(LWR),chalkgrainrate(CGR),chalkinessdegree(CD),gelatinizationtemperature(GT),amylosecontent(AC)andgelconsistency(GC)ofheadrice.ThreemappingmethodsemployedwerecompositeintervalmappinginQTLMapper2.0softwarebasedonmixedlinearmodel(MCIM),inclusivecompositeintervalmappinginQTLIciMapping3.0softwarebasedonstepwiseregressionlinearmodel(ICIM)andmultipleintervalmappingwithregressionforwardselectioninWindowsQTLCartographer2.5basedonmultipleregressionanalysis(MIMR).ResultsshowedthatfiveQTLswithadditiveeffect(A-QTLs)weredetectedbyallthethreemethodssimultaneously,twobytwomethodssimultaneously,and23byonlyonemethod.FiveA-QTLsweredetectedbyMCIM,ninebyICIMand28byMIMR.ThecontributionratesofsingleA-QTLrangedfrom0.89%to38.07%.AlltheQTLswithepistaticeffect(E-QTLs)detectedbyMIMRwerenotdetectedbytheothertwomethods.FourteenpairsofE-QTLsweredetectedbybothMCIMandICIM,and142pairsofE-QTLsweredetectedbyonlyonemethod.Twenty-fivepairsofE-QTLsweredetectedbyMCIM,141pairsbyICIMandfourpairsbyMIMR.ThecontributionratesofsinglepairofE-QTLwerefrom2.60%to23.78%.IntheXiu-BaoRILpopulation,epistaticeffectplayedamajorroleinthevariationofGLandCD,andadditiveeffectwasthedominantinthevariationofLWR,whilebothepistaticeffectandadditiveeffecthadequalimportanceinthevariationofCGR,AC,GTandGC.QTLsdetectedbytwoormoremethodssimultaneouslywerehighlyreliable,andcouldbeappliedtoimprovethequalitytraitsinjaponicahybridrice.
简介:明确稻瘟病抗性与水稻主要农艺性状的相关性,对于抗源改良及新抗源的创制具有指导意义。以重组自交系C55为材料,连续2年在水尾病圃通过田间自然诱发稻瘟病考察C55各家系的颈瘟病情指数,在泸县考察C55各家系的主要农艺性状,分析颈瘟病指与主要农艺性状的相关性;选取田间最高颈瘟病指<23.0%的15个家系组成抗病组,选取单株粒重>34.0g的15个家系组成感病组,通过t检验比较2组间主要性状的差异显著性。结果表明:播始天数、单株穗数及株高与颈瘟病指的简单相关呈极显著负相关(P<0.01),穗实粒数、穗着粒数、千粒重及单穗重与颈瘟病指的简单相关呈极显著正相关(P<0.01);播始天数、株高及结实率与颈瘟病指的偏相关呈极显著负相关(P<0.01),单穗重与颈瘟病指的偏相关呈极显著正相关(P<0.01)。抗、感2组间颈瘟病指、穗实粒数、穗着粒数、单穗重及单株粒重的差异达5%或1%显著水平。单株产量高的家系其抗瘟性相对弱,15个抗病组家系可作为抗源利用,并对抗源改良方式进行了讨论。
简介:IL-16isaligandandchemotacticfactorforCD4+Tcells.IL-16inhibitstheCD3mediatedlymphocyteactivationandproliferation.TheeffectsofIL-16onthetargetcellsaredependentonthecelltype,thepresenceofco-activatorsetc.TounderstandtheregulationfunctionandmechanismofIL-16ontargetcells,weuseda130a.a.recombinantIL-16tostudyitseffectsonthegrowthofJurkatTleukemiacellsinvitro.WefoundthattherIL-16stimulatedtheproliferationofJurkatcellsatlowdose(10^-9M),butinhibitedthegrowthofthecellsathigherconcentration(10^-5M).Resultsshowedthat10^-5MofrIL-16treatmentinducedanenhancedapoptosisinJurkatcells.ThetreatmentblockedtheexpressionofFasL,butup-regulatedthec-mycandBidexpressioninthecells.Pre-treatmentofPKCinhibitororMEK1inhibitormarkedlyincreasedordecreasedtherIL-16inducedgrowth-inhibitingeffectsonJurkatcells,respectively.TheresultssuggestedthattherIL-16mightbearegulatorforthegrowthorapoptosisofJurkatcellsatadose-dependentmanner.Thegrowth-inhibitingeffectsofrIL-16mightbeFas/FasLindependent,but,associatedwiththeactivationofPKC,up-regulatedexpressionofc-MycandBid,andtheparticipationoftheERKsignalpathwayinJurkatcells.
